C-type lectin domain family 12 member B (CLEC12B), also known as Macrophage antigen H, is an inhibitory receptor that negatively regulates immune and non-immune functions. Upon phosphorylation, it recruits SH2 domain-containing PTPN6 and PTPN11 phosphatases via its ITIM motif to antagonize activation signals. It limits myeloid cell activation during infection or inflammation, protects target cells against natural killer cell-mediated lysis, and negatively regulates melanocyte cell cycle and differentiation through STAT3 inactivation.
