    Protein ENL, encoded by the MLLT1 gene, is a chromatin reader component of the super elongation complex (SEC) that enhances RNA polymerase II transcription by reducing polymerase pausing. It specifically binds acetylated and crotonylated histones, with a preference for crotonylated histones such as H3K27cr and H3K9cr20. ENL plays a critical role in acute myeloid leukemia by recognizing acetylated histones via its YEATS domain, thereby driving oncogenic gene transcription. The protein is localized to the nucleus, underscoring its involvement in nuclear epigenetic regulation and gene expression control.
